ABSTRACT

Aims: SA (Salicylic acid) is a key regulator for sugar metabolic pathway, but the regulating function on sugar metabolism in tomato seedlings is imperfect. In this study, tomato seedlings were sprayed with SA, and then soluble sugar contents, enzyme related to sugar metabolism levels, gene expressions were determined. Study Design: In order to analyze the mechanism of SA in sugar metabolic pathway, tomato seedlings were sprayed with SA, and then soluble sugar contents, enzyme related to sugar metabolism levels, gene expressions were determined. Place and Duration of Study: College of Biological Science and Technology, between December 2021 and May 2022. Methodology: The expressions of genes related to sugar metabolism in tomato were measured by qRT-PCR. The contents of soluble sugar in tomato seedings were measured with HPLC (High performance liquid chromatography). Results: The sugar metabolism in tomato seedlings was affected by SA treatment for 6 h. The contents of fructose, glucose and sucrose were improved in tomato seedlings by SA treatment. Between 6 to 24 h, enzymatic activities of SlAI, SlNI and SlSS were higher than those in controls, while there was little change in enzyme activity of SISPS. Conclusion: The contents of soluble sugar, enzyme activities related to sugar metabolism had changed considerably by SA treatment. Especially from 6 h to 24 h after SA treatment, the contents of fructose and glucose were significantly higher than those of the control group, and the changes of sugar contents were mainly related to the increases of enzyme activities of AI, NI and SS, suggesting that SA treatment mainly affected the activities of SlAI, SlNI and SlSS to promote the accumulations of soluble sugars.

The inefficient seedling production and the slow development of saplings, combined with the impact of the extractivism and the high rate of predation of the pyrenes, suggest the low recruitment rate of the species observed, in natural conditions. This type of data is one of the important tools for creating guidelines for the species conservation. Therefore, we suggest considering the reclassification of B. purpurascens as a Critically Endangered species in the Official List of Threatened Brazilian Species of Extinction.

A capacidade de produção de plântulas ineficiente e o lento desenvolvimento das mudas, combinados com o impacto do extrativismo e a alta taxa de predação dos pirênios, sugerem a baixa taxa de recrutamento da espécie observada, em condições naturais. Esse tipo de dado é uma das ferramentas importantes para a criação de diretrizes para a conservação de espécies. Portanto, sugerimos considerar a reclassificação de B. purpurascens como espécie Criticamente Ameaçada na Lista Nacional Oficial de Espécies da Flora Brasileira Ameaçadas de Extinção.

ABSTRACT: Macronutrient suppression is one way to identify which chemical elements is the most important in the early development of seedlings of forest species. This study evaluated the initial growth of Eucalyptus benthamii seedlings through morphological and physiological variables, submitted to doses of N, P and K, and liming. The experimental design to install the experiment was completely randomized, with a 2x9 factorial scheme, in which levels of factor "A" referred to absence and presence of liming and levels of factor "B", to the different treatments of NPK. Morphological variables of SD (stem diameter) and H (height) and the physiological variables of A (photosynthesis), E (transpiration), gs (stomatal conductance), Ci/Ca (relationship between intercellular and atmospheric CO2 concentration) and WUE (water use efficiency) were measured. For morphological variables, the absence of N directly affected the means and there was a direct relationship between increase of the dose and increase of SD and H. There was a positive relationship of P mainly in relation to shoot dry matter content (SDMC) and root dry matter content (RDMC). For the physiological variables, there was no direct response with the doses of N or P, with K being the element that most influenced the variables, especially when limestone was applied, resulting in higher averages for photosynthesis, transpiration and stomatal conductance. With the purpose of meeting the morphological and physiological demands, the recommendation for cultivation of Eucalyptus benthamii under the conditions of this study is the doses 150-200-150, without liming application.


RESUMO: A supressão dos macronutrientes é uma das maneiras de identificar qual ou quais dos elementos químicos é o mais importante no desenvolvimento inicial das mudas de espécies florestais. O estudo objetivou avaliar o crescimento inicial de mudas de Eucalyptus benthamii por meio de variáveis morfológicas e fisiológicas, submetidas a doses de N, P e K, e calagem. O delineamento experimental para instalação do experimento foi em delineamento inteiramente casualizado, com esquema fatorial 2x9, em que os níveis do fator "A" se referiram a ausência e a presença de calagem e os níveis do fator "B" aos diferentes tratamentos de NPK. Variáveis morfológicas de DAC (diâmetro a altura do colo) e H (altura) e as variáveis fisiológicas de A (fotossíntese), E (transpiração), gs (condutância estomática), Ci/Ca (relação entre a concentração intercelular e atmosférica de CO2) e WUE (eficiência no uso da água) foram determinadas. Para as variáveis morfológicas, a ausência do N afetou diretamente as médias, havendo uma relação direta entre o aumento da dose com o aumento das variáveis. Há uma relação positiva do P principalmente em relação a MSPA e MSPR. Para as variáveis fisiológicas, não houve resposta direta com as doses de N ou P, sendo o K o elemento que mais influenciou as variáveis, principalmente quando o calcário foi aplicado, resultando em maiores médias para fotossíntese, transpiração e condutância estomática. Com o objetivo de atender às demandas morfológicas e fisiológicas, a recomendação para o cultivo de Eucalyptus benthamii nas condições deste estudo é a dose 150-200-150, sem aplicação de calagem.

ABSTRACT

The hydroponic culture test method was used to study the physiological and biochemical responses of Paulownia fortunei seedlings under Zn stress, Cd stress, and combined Zn and Cd stress as well as changes in the enrichment and transfer characteristics of heavy metals. Under single and combined heavy metal stress, the biomass, plant height, and peroxidase (POD) activity of P. fortunei decreased as the treatment concentration increased. Combined Zn and Cd affected adversely plant height and biomass. As the concentration of Zn increased when applied alone, the chlorophyll content and catalase (CAT) activity of P. fortunei first increased and then decreased, the superoxide dismutase (SOD) activity increased, and the aboveground malondialdehyde (MDA) content first decreased and then increased. As the concentration of Cd increased when applied alone, chlorophyll content and CAT activity increased, and SOD activity and aboveground MDA content first increased and then decreased. Under both Cd and Zn, the physiological response was more complex. Cd in the seedlings of P. fortunei was concentrated in the root. In contrast, Zn was concentrated in the upper part of the ground, and its transfer coefficient was greater than 1.00. Thus, the addition of Zn promotes the transfer of heavy metals to the above-ground portions of plants. Generally, P. fortunei can effectively promote ecological restoration under complex forms of heavy metal pollution.

ABSTRACT

Yield and longevity of yellow passion fruit have been reduced by diseases such as the bacterial spot caused by Xanthomonas axonopodis pv. passiflorae. Genetic resistance has been confirmed as the most efficient and economical correct option to minimize this disease problem. Aiming at it, the objective of this research was to evaluate the incidence, severity and progression of the disease in 12 genotypes of sour passion fruit, in seedling stage in nursery greenhouse after inoculation of Xanthomonas axonopodis pv. passiflorae. The inoculation was performed with an isolate collected in the Pipiripau Rural Nucleus, Brasilia-DF, named UnB-1397 (1x106 CFU/mL), through induction of injuries. There were performed four assessments, with interval of 7 days except the first which was performed 11 days after inoculation. The incidence was estimated by the percentage of plants affected. To evaluate the severity, it was used the diagrammatic scale validated by Costa et al. (2018), with adaptations, using the measurement of the affected area by necrotic lesions on the leaf. All genotypes were susceptible to bacteriosis, 5 being considered moderately susceptible: F1BRS Pérola do Cerrado x Rosa Intenso, Mar20#21, Mar20#15b, Mar20#24xMar20#40 and FB200PL4R2 x Mar20#2005, with a mean of severity ranging from 11 to 25% of injured area in leaves.


A produtividade e a longevidade dos pomares de maracujazeiro-azedo têm sido comprometidas em razão de doenças como a bacteriose, causada por Xanthomonas axonopodis pv. passiflorae. A resistência genética tem se confirmado como a opção mais eficiente e econômica para minimizar tal problema. Dessa forma, o objetivo deste trabalho foi avaliar a reação de 12 genótipos de maracujazeiro, em fase de mudas, sob cultivo protegido, à Xanthomonas axonopodis pv. passiflorae. A inoculação com isolado denominado UnB-1397 (1x106 CFU/mL),coletado no Núcleo Rural de Pipiripau, Brasília-DF, se deu pela indução de ferimentos. Foram realizadas 4 avaliações, com intervalo de 7 dias, sendo a primeira avaliação realizada 11 dias após a inoculação. A incidência foi estimada pela porcentagem de plantas afetadas. Para avaliação da severidade, foi utilizada escala diagramática validada por Costa et al. (2018), com adaptações, utilizando-se a mensuração da área foliar atingida por lesões nas folhas. Todos os genótipos se mostraram suscetíveis à bacteriose, sendo 5 considerados moderadamente suscetíveis: F1 BRS Pérola do Cerrado x Rosa Intenso, Mar20#21, Mar20#15b, Mar20#24 x Mar20#40 e FB200PL4R2 x Mar20#2005, apresentando uma média de severidade que variou de 11 a 25% de área ou tecidos foliares lesionados.

Objective::To investigate the effects of plant-soil feedback on secondary metabolites in roots, stems and leaves of Acanthopanax senticosus seedlings. Method::One-year-old seedlings of A. senticosus were planted in the soil where no A. senticosus had been planted before (group 1), soil where A. senticosus had been planted for 3 years (group 2), and soil where A. senticosus had been planted for many years in the greenhouse pot experiment, and the secondary metabolites of its roots, stems and leaves were then analyzed. Result::L-Phenylalanine, 3, 4-dihydroxybenzoic acid, syringin, chlorogenic acid, caffeic acid, eleutheroside E, isofraxidin, rutin, hyperoside, and quercetin had significant differences in leaves and roots of A. senticosus seedlings in the soil of group 3, but there was no significant difference in chlorogenic acid and eleutheroside E in stem. Eleutheroside E, isofraxidin, rutin and hyperoside were not detected in the leaves of seedlings planted in group 3.Most of the secondary metabolites in the roots of A. senticosus seedlings showed positive feedback, while in the stems of Acanthopanax senticosus seedlings, caffeine, A. senticosus glycosides, hypericin and quercetin showed negative feedback, and most of the secondary metabolites in the leaves of A. senticosus seedlings showed positive feedback. Conclusion::The plant and soil showed different feedback in different parts of the growth process of A. senticosus seedlings, and the soil where A. senticosus had not been planted was more advantageous to the secondary metabolites of A. senticosus seedlings. The results of the study provide a basis for the study of the effect of plant-soil feedback on the A. senticosus, and provide the theoretical basis and technical support for the artificial cultivation of A. senticosus.

ABSTRACT

Abstract Passion fruits species presents a larger economic importance for in natura consumption and industry. The main way to propagate these species is by seeds; however, the aril presence and environmental conditions may inhibit the seeds germination. In this sense, the objective of this study was to evaluate the effect of aril removal methods and light conditions on P. edulis var. flavicarpa seeds quality and germination. There were used five methods to remove the seeds aril: (1) friction on steel mesh sieve for 5 minutes; (2) friction with coarse sand on steel mesh sieve for 5 minutes; (3) immersion in lime solution at 10% (w/v), for 30 minutes; (4) fermentation in sugar solution at 10% (w/v), during 48 hours; and (5) natural pulp fermentation, during five days. The seeds were submitted to germination test at darkness and lightness, as well as, to length seedlings and emergence test. Regardless the aril removal method, the germination percentage was lower at lightness than darkness highlighting the natural pulp fermentation treatment, which presented 86% of seeds germinated. On the other hand, the aril removal methods did not affect the seedlings emergence, with emergence percentages ranged from 88 to 94%.


Resumo As espécies de maracujá apresentam grande importância econômica, tanto para o consumo in natura, quanto para a indústria. A principal maneira de propagar essas espécies é por sementes; contudo, a presença de arilo e condições ambientais pode inibir a germinação. Nesse sentido, objetivou-se estudar o efeito de métodos de remoção do arilo e luminosidade na qualidade e germinação das sementes de Passiflora edulis var. flavicarpa. Cinco métodos foram empregados para a remoção do arilo: (1) fricção em peneira de malha de aço por cinco minutos; (2) fricção com areia grossa em peneira de malha de aço por cinco minutos; (3) imersão em solução de cal a 10% (m/v), por 30 minutos; (4) fermentação da polpa em solução de açúcar a 10% (m/v), por 48 horas; e (5) fermentação natural da polpa, por cinco dias. As sementes foram submetidas ao teste de germinação no escuro e no claro, teste de comprimento de plântulas e teste de emergência. Independente do método de remoção do arilo, a porcentagem de germinação no claro apresentou-se menor em comparação ao escuro, com destaque para o tratamento de fermentação natural da polpa, o qual apresentou 86% de sementes germinadas. Por outro lado, os métodos de remoção do arilo não afetaram a emergência das plântulas, com porcentagem de emergência variando entre 88 a 94%.

ABSTRACT

Salinity is one of the most limiting environmental factors for plant growth in semi-arid regions.Excessofsalts reduces water potential, causes physiological drought and can influence germination and initial growth of plants. The objective of this work was to study physiological and metabolic changes in seeds and seedlings of pumpkin (Cucurbita pepo cultivars Caserta and Redonda and C. maxima cv. Coroa)in a saline condition of salt stress. Thus, we it was quantified germination parameters; seedlings growth; seeds and seedlings respiration; degradation of total soluble sugars, reducing sugars and total proteins in cotyledons and the activity ascorbate peroxidase (APX), glutathione-S-transferase (GST) and catalase (CAT) during germination and initial growth of the pumpkin species studied in saline conditions. The experimental design was totally randomized in a factorial scheme 6x3, with six electrical conductivities (0; 2; 4; 6; 8 e 10 dS.m-1) and three cultivars (Redonda, Caserta and Coroa). Salinity did not influence the rate of seed respiration during the germination, although seedlings respiration decreased, as well as its vigour. Total sugars were degraded, while there was accumulation of reducing sugars and proteins in cotyledons of germinating seeds in salt solutions. Increased electrical conductivity reduced APX activity in the embryo and CAT in cotyledons, but did not influence GST. Pumpkin seedlings showed low tolerance to salt stress.


A salinidade é um dos fatores limitantes para o crescimento das culturas nas regiões áridas e semiáridas. O excesso de sais provoca uma redução no potencial hídrico e causa seca fisiológica e influencia na germinação e crescimento inicial de plântulas Objetivou-se estudar possíveis alterações fisiológicas e metabólicas em sementes e plântulas de abóboras (Cucurbita pepo cultivars Caserta and Redonda and C. maxima cv. Coroa), Sendo assim, avaliou-se em sementes e plântulas os parâmetros da germinação (porcentagem e velocidade); crescimento de plântulas; respiração de sementes e plântulas; degradação de açúcar solúveis totais, açúcar redutores e proteínas totais nos cotilédones e atividade das enzimas ascorbato peroxidase (APX), glutationa-S-transferase (GST) e catalase (CAT) durante a germinação e crescimento inicial das cultivares de abóbora em soluções salinas. O delineamento experimental foi inteiramente casualizado em esquema fatorial 6x3, com seis condutividades elétricas (0; 2; 4; 6; 8 e 10 dS.m-1) e três cultivares (Redonda, Caserta e Coroa). A salinidade não influenciou a taxa de respiração das sementes durante a germinação, no entanto, a respiração das plântulas diminuiu, assim como o vigor das plântulas. Houve degradação dos açúcares totais e acúmulo de açúcares redutores e proteínas em cotilédones de sementes submetidas a soluções salinas. O aumento da condutividade elétrica induziu redução da atividade de APX no embrião e de CAT nos cotilédones, mas não influenciou GST. Cultivares de aboboras apresentaram baixa tolerância ao estresse salino.

ABSTRACT

Erythroxylum pauferrense is an endemic understory plant species of the Northeast Region of Brazil. The species is of great importance to the region and so ecophysiological studies are needed for its preservation. The objective of the present study was to determine the best substrates and temperatures for testing germination and seed vigor of E. pauferrense. An experiment was performed comprising a completely randomized design in a 5 x 4 factorial scheme, with five temperature regimes (20, 25, 30, 35°C constant and 20-30°C alternating) and four types of substrates (paper, vermiculite, sand and commercial substrate). The following characteristics were evaluated: germination percentage, first germination count, germination speed index, mean germination time, seedling length and dry mass (root and shoot). Paper and vermiculite substrates combined with constant temperatures of 20, 25, 30 and alternated between 20-30°C, provide greater seed germination and vigor while 35°C reduces seed physiological quality.

ABSTRACT

Objective: To study the seeds shapes and germination characteristics, construct aseptic seedlings and callus cultural system, and provide a basis for the rapid propagation and secondary metabolic regulation of Rheum palmatum. Methods: Ten batches of R. palmatum seeds were subjected to characteristic analyses from the aspects of size, purification, weights per thousand seeds, seeds vigor, germination rates, and germination energy. The optimum disinfection system for aseptic seedlings and the optimum hormone ratio for inducing callus were screened by orthogonal test. The content of ten active components in aseptic seedlings and calli were primarily evaluated by HPLC analysis. Results: There was no significant difference in the appearances of the ten batches of R. palmatum seeds from different regions. The content of moisture, seeds vigor, germination rates, and germination energy differed apparently. The germination characteristics in Hezheng and Weiyuan counties were the best. The best disinfection group for aseptic seedlings was the combination of 75% ethanol for 30 s with 10% hydrogen peroxide for 15 min. The optimum hormones for callus induction were 6-BA (1.0 mg/L) + KT (2.0 mg/L) + NAA (1.5 mg/L). Seeds treated with different disinfectants had no significant effect on the content of ten components in germinated aseptic seedlings (P > 0.05). Seven active components were detected in callus, which was significantly lower than that in aseptic seedlings. And the content of chrysophanol-8-O-glucoside was the highest in the callus. Conclusion: The seed characteristics from Hezheng and Weiyuan counties in Gansu Province were excellent by analyses of weights per thousand seeds, seeds vigor, germination rates, and germination energy. The aseptic seedlings and callus cultural system of R. palmatum were successfully established, laying a solid foundation for further study.

ABSTRACT

Objective:Stems,petioles,stem sections with axillary and leaves of Scrophularia ningpoensis were taken as the material in vitro to screen out the suitable plantlet regeneration system and optimal exercising seedling conditions. Method:Different explants,hormones and concentrations on the induction and proliferation of cluster bud were studied by L16(45) orthogonal test. One factor analysis of variance (ANOVA) was made on the induction of adventitious buds rooted with different concentrations of hormones. At the same time,different substrates,watering cycles and transition modes were selected to optimize key technologies of exercising seedlings of S. ningpoensis. Result:Stem sections with axillary was the best explant,which was followed by stems,leaves and petioles. The suitable media for the induction of adventitious buds was MS+6-BA 0.5 mg·L-1+NAA 0.2 mg·L-1,with the induction rate of 100.0% and the proliferation multiple of 9.84.The suitable media for root induction was 1/2 MS+IBA 0.2 mg·L-1,with the rooting rate of 100.0% and the number of roots of 39.45.For matrix,they were transplanted with nutrient soil,vermiculite and perlite (5:2:1) as the media,to keep proper matching of fertility,permeability and water retention. The container seedlings can grow well,and the survival rate was more than 95% when they were watered every 2 days,the acclimatization of plantlets took 20 days indoor and 10 days in shaded greenhouses. Conclusion:The clonal propagation system of S. ningpoensis was established to provide an effective way for the efficient,rapid and steady plantlet regeneration,the breeding of high-quality seedlings and the suitable exercising seedling conditions of S. ningpoensis.

ABSTRACT

The aim of the study is to explore exogenous S3307 on alleviating low-temperature stress of coix seedlings. The coix cultivar, "No 5 Yiliao", was selected as the plant material, through nutrient solution cultivating in greenhouse, the effect of different S3307 concentrations(1, 3, 5, 7, 9 mg·L~(-1)) on coix seedlings traits and physiological indicators were explored under low-temperature stress. The results showed, under low-temperature 5 mg·L~(-1) S3307 could significantly increase coix seedlings stem diameter and biomass, which stem diameter and above-ground biomass, low-ground biomass separately were enhanced 11.90%, 13.59%, 10.99%. Leaf width and lateral root number separately were enhanced 7.63%, 37.52%. Meanwhile, addition of 5 mg·L~(-1) S3307 could significantly reduce relative conductivity and MDA, separately being reduced 23.33%, 17.42% compared to CKL. S3307 could also significantly increase soluble sugar and proline content, which leaf soluble sugar and proline content separately were enhanced 17.16%, 11.87%, which root soluble sugar and proline content separately were enhanced 20.00%, 33.42%. Additionally, S3307 could alleviate the cells destroy in ultra-structure level by improving cell membrane structure and chloroplast capsule layer structure. 5 mg·L~(-1) S3307 could enhance the low temperature tolerance of coix seedlings by regulating the growth and physiological indexes, and thus alleviate the damage caused by low-temperature to the coix seedlings.

ABSTRACT

To develop more active LTR retrotransposons in Phyllostachys edulis, a Ph. edulis LTR retrotransposon (Ph-LTR2) was identified, and the expression pattern of the transposon under stress was systematically analyzed. Ph-LTR2 transposon is 6 030 bp in length and belongs to the Reina subfamily in the Ty3-Gypsy family. With the similarity of 96.41% of both LTR sequences, the Ph-LTR2 transposon inserted the moso bamboo genome about 61.92 thousand years ago. There are 5 copies identified in the genome. The Ph-LTR2 transposon domain includes GAG (gag protein) protein domain, PR (Proteases) protein domain, RT (Reverse transcriptase) protein domain, RH (Ribonuclease H) protein domain, INT (Integrase) protein domain and CHR (Chromatin organization modifier) protein domain. The expression patterns of INT, RT and RH were detected by real-time quantitative PCR. The three domains were found to have specific expression patterns at different tissues of the bamboo. Under the conditions of low/high temperature, methylation inhibitors treatments, irradiation and high salt stress, transcription levels of the three domains of the Ph-LTR2 transposon increased with different degrees. Specifically, after treatment with low/high temperature and methylation inhibitors, the transcription level was up-regulated; after low dose radiation treatment and low concentration of salt solution treatment, the transcription level was also increased, but the expression level decreased with increasing dose of radiation and concentration of salt solution. These results indicate that the expression pattern of the Ph-LTR2 transposon responds to the changes of the external environment, but the exact mechanism is not yet known. The results of this study laid a certain theoretical foundation for the development of the genetic tool based on Ph-LTR2 transposons.

ABSTRACT

As an important signal molecule, extracellular ATP(eATP) can regulate many physiological and biochemical responses to plant stress. In this study, the regulation of extracellular ATP(eATP) on chlorophyll content and chlorophyll fluorescence parameters of Angelica sinensis seedlings were studied under drought and low temperature stress. The results showed that all the chlorophyll content, the actual photochemical efficiency [Y(Ⅱ)], the electron transfer rate(ETR), the photochemical quenching coefficient(qP and qL) of A. sinensis leaves were significantly decreased under drought and low temperature stress, respectively. At the same time, non-photochemical quenching(NPQ and qN) were also all significantly increased, respectively. The application of eATP alleviated the decrease of chlorophyll content, Y(Ⅱ), ETR, qP and qL of A. sinensis leaves under drought and low temperature stress, and eliminated the increase of qN and NPQ. The results indicated that eATP could effectively increase the open ratio of PSⅡ reaction centers, and improve the electron transfer rate and light energy conversion efficiency of PSⅡ of A. sinensis leaves under drought and low temperature stress. It is beneficial to enhance the chlorophyll synthesis and the adaptability of PSⅡ about A. sinensis seedlings to drought and low temperature stress.

ABSTRACT

The pata-de-vaca (Bauhinia divaricata L.) tree is a species widely distributed in Brazil, it has a high ornamental and economic value. Its leaves are used in renal inflammations, such as diuretic, hypoglycemic, being considered a medicinal plant of popular use. The research was carried out in a greenhouse belonging to the Laboratório de Ecologia Vegetal, at the Centro de Ciências Agrárias (CCA), from Universidade Federal of Paraíba (UFPB). The objective was to evaluate the morphophysiological characteristics of B. divaricata plants at different water regimes. The experimental design was completely randomized with five water treatments 1 (100% control), 2 (80%), 3 (60%), 4 (40%) and 5 (20%) of the container capacity. The height, number of leaves and stem diameter were evaluated weekly. At the end of the experiment the plants had their organs separated and taken to the greenhouse to obtain the dry matter of the leaves, stem, roots and total dry matter, and also, biomass allocation in the leaves, stem and roots. Plant height did not differ statistically between the treatments. The number of leaves, diameter, leaf, stem and root allocation and root and shoot dry matter were higher under water availability (100% of container capacity). For the production of B. divaricata L. seedlings water regime should be 100% and 80% of the container capacity, but the seedlings grow satisfactorily.


A pata-de-vaca (Bauhinia divaricata L.) é uma espécie arbórea, amplamente distribuída no Brasil, de alto valor ornamental e econômico. Suas folhas são utilizadas nas inflamações renais, como diuréticas, hipoglicemiantes, sendo considerada uma planta medicinal de uso popular. A pesquisa foi desenvolvida em casa de vegetação pertencente ao Laboratório de Ecologia Vegetal, Centro de Ciências Agrárias (CCA), Universidade Federal da Paraíba (UFPB). O Objetivo foi avaliar as características morfofisiológicas em plantas de Bauhinia divaricata em diferentes regimes hídricos. O delineamento experimental foi inteiramente casualizado com cinco tratamentos hídrico 100%, 80%, 60%, 40% e 20% da capacidade de pote. Avaliou-se, semanalmente a altura, número de folhas e diâmetro do caule. Ao final do experimento as plantas tiveram seus órgãos separados e levados à estufa para obtenção da matéria seca das folhas, caule, raízes e massa seca total, e ainda, alocação de biomassa nas folhas, caule e raízes. A altura de plantas não diferiu estatisticamente entre os tratamentos, o número de folhas, diâmetro, alocação de folhas, caules e raízes e massas seca da raiz e parte aérea foram maiores sob maior disponibilidade de água (100% da capacidade do recipiente). Para a produção de mudas de B. divaricata L. o regime hídrico deve ser de 100% e 80% da capacidade do recipiente, porém as mudas crescem satisfatoriamente.

ABSTRACT

Aims: An experiment was conducted to investigate the effect of a wide range of temperatures on the growth and physiology of Theobroma cacao, to study the differences between night and day temperatures and to determine the optimum temperature for the cocoa growth. Study Design: The experiment used five combinations of night and day temperatures (18°C and 30°C [18N30N], 18°C and 36°C; [18N 36D], 24°C and 24°C [24N24D], 24°C and 30°C [24N30D] and 24°C and 36°C [24N36D]) using complete randomized design (CRD). Place and Duration of Study: Crops and Environment Laboratory University of Reading and International Cocoa Quarantine Centre, between 23rd May 2016 and 25th July 2016. Methodology: The cocoa seedlings were put into five growth cabinets with five different night and day temperatures combinations (18°C and 30°C, 18°C and 36°C, 24°C and 24°C, 24°C and 30°C, 24°C and 36°C) for two months (63 days) under controlled environment condition where the relative humidity and vapor pressure deficit were controlled. Destructive harvest data was taken at end of the experiment which included fresh weight, dry weight, leaf area and root weight. Non-destructive measurements were height of the plant, photosynthetic rate, chlorophyll fluorescence and total chlorophyll content. Results: Treatment 24N30D have the best growth and treatment 24N36D had the lowest growth performances compared to other treatments. Conclusion: The growth was not only dependent on the day temperature, but also on the night temperature. A large gap between night and day temperatures (DIF) reduced the cocoa growth. The result also showed the optimum temperature amongst those studied for cocoa growth is the combination of 24°C night temperature and 30°C day temperature.

Abstract Aspergillus sp., Fusarium sp., and Ramularia sp. were endophytic fungi isolated from Rumex gmelini Turcz (RGT), all of these three strains could produce some similar bioactive secondary metabolites of their host. However the ability to produce active components degraded significantly after cultured these fungi alone for a long time, and were difficult to recover. In order to obtain more bioactive secondary metabolites, the co-culture of tissue culture seedlings of RGT and its endophytic fungi were established respectively, and RGT seedling was selected as producer. Among these fungi, Aspergillus sp. showed the most significant enhancement on bioactive components accumulation in RGT seedlings. When inoculated Aspergillus sp. spores into media of RGT seedlings that had taken root for 20 d, and made spore concentration in co-culture medium was 1 × 104 mL-1, after co-cultured for 12 d, the yield of chrysophaein, resveratrol, chrysophanol, emodin and physcion were 3.52-, 3.70-, 3.60-, 4.25-, 3.85-fold of the control group. The extreme value of musizin yield was 0.289 mg, which was not detected in the control groups. The results indicated that co-culture with endophytic fungi could significantly enhance bioactive secondary metabolites production of RGT seedlings.

The genus Citrus contains numerous fresh and processed fruit cultivars that are economically important worldwide, many genotypes are amenable to somatic embryogenesis, which became a key regeneration pathway in many experimental approaches of cultivar improvement. in this objective We aime at studying the effects of various culture media on the induction and the development of citrus somatic embryos.Callus cultures were initiated from the infertlized ovules of six varieties of mandarin (Anana, Lee, Murcott, Ortanique, Temple, and Wilking) within 3 media: MT (Murashig and Tuker, 1969) without hormones, MT + 1 mg/l BAP, MT + 1 mg/l Kinetin, the experiments show a highly significant effect (P < 0.001) of the culture media and genotype. No reactivity was observed on the MT environment in the absence of growth regulator, while the culture media MT in addition to 1 mg/l BAP gave the best results of induction of embryogenic callus induction. The induction of somatic embryogenesis was obtained on MT media without hormones. For the plantlets regeneration the favorable media was MT without hormones or added to ANA and active coal.
